What it is
Winter grafted the complementarity-determining regions (CDRs) of a mouse antibody into a human antibody framework. The result keeps the specificity but triggers much less immune response, and it solved the limitation of murine hybridoma antibodies.
Winter's 1986 patent and the first clinical application, Campath-1H (Riechmann et al. 1988), came from the same group. Chimaeric antibodies (Morrison, 1984) were an earlier step.
What it led to
- therapeutic antibodies such as alemtuzumab (Campath-1H), the first humanised, and trastuzumab
- multi-billion therapeutic antibody market
- fully human phage antibodies
